4 million Windows 8 upgrades sold already, says Ballmer

Microsoft CEO Steve Ballmer announced at the company's Build conference today?that more than 4 million Windows 8 upgrades had been sold since?the operating system's official launch on Friday. He also said "tens of millions of copies" had been shipped to partners.

Ballmer said?yesterday?that "preliminary demand" was higher for Windows 8 than it was for Windows 7, but offered no numbers to back up that claim at the time. The 4 million upgrades he announced today give some idea of the OS's success, but putting the numbers in context is actually quite complicated.

Aside from yesterday's remark, Windows 8 sales have not been compared to Windows 7's ? that's?not surprising considering the OS has only been available for a few days. Even when Windows 7 was moving?enough copies?to be considered the fastest-selling OS in history, Microsoft waited several weeks before announcing how sales compared to those of Windows Vista. So it will likely be some time before more solid numbers are announced.

Singling out upgrades complicates things further: People have been registering and qualifying for these reduced-price copies of Windows 8 since June, when new PCs began to ship with the guarantee of a $15 upgrade to the new OS upon launch.

Four million may or may not be considered a large number, depending on how it is interpreted in light of total PC sales. Numbers from Gartner put worldwide PC sales in the third quarter of 2012 (which roughly corresponds to the pre-order and upgrade period for Windows 8) at about 87.5 million. Theoretically, all those PCs qualify for the $15 upgrade. The 4 million represents?a 4.5 percent conversion rate for new PCs in the first ?week ? that sounds low, but these numbers are so early that few conclusions can be drawn.

As for the tens of millions of copies shipped to partners, that comes as no surprise. PC makers are banking on the novelty of Windows 8 to sell dozens of new, touch-enabled PCs and tablets, and in order to ship with Windows 8, manufacturers like Lenovo and Acer will have ordered licenses by the million?? although they will likely offer Windows 7 for some time to come as a budget option.

There are no firm take-aways from Ballmer's numbers, which were likely shared in order to whet the appetite of shareholders, developers and reporters, all eager to know the results of Microsoft's risky experiment in redesigning Windows. The next few weeks should bring more intelligible numbers and the opportunity to compare them, apples?to?apples, with Windows 7's sales.

Tunisia: Reported consulate suspect arrested

TUNIS, Tunisia (AP) ? A Tunisian man who was arrested in Turkey this month with reported links to the attack on a U.S. consulate in Libya has been returned to Tunisia and is facing terrorism charges, his lawyer said Wednesday.

Ali Harzi was repatriated to Tunisia on Oct. 11 by authorities in Turkey, and a judge issued his arrest warrant, lawyer Ouled Ali Anwar told The Associated Press. He said his client was told by a judge Tuesday that he has been charged with "membership of a terrorist organization in a time of peace in another country."

A person who saw Harzi's court dossier told The Associated Press that the file links him to the Sept. 11 attack on the U.S. consulate in Benghazi that left Ambassador Christopher Stevens and three other Americans dead.

He said Harzi is one of two Tunisians reportedly arrested Oct. 3 in Turkey when they tried to enter the country with false passports. The person spoke on the condition of anonymity because he was not authorized to release the information. Harzi's alleged role in the attack is not clear.

Anwar denied there was any evidence that Ali was implicated in the attacks. He added his client was not using a fake passport, saying he was a "scapegoat to satisfy the Americans."

The charge against Harzi is punishable by six to 12 years in prison, according to the provisions of the anti-terrorist law in force in Tunisia since 2003.

Earlier this month, Defense Secretary Leon Panetta said the U.S. has been looking into the arrests of two Tunisian men being detained in Turkey reportedly in connection with the attacks. The State Department in Washington had no further comment on Wednesday.

A U.S. intelligence official was cautious about the Tunisian arrest, saying that the Tunisians have so far not allowed American officials to interview the suspect, so the U.S. is not yet certain how directly he is connected to the attack.

The suspect has ties to both Ansar al-Shariah and Al Qaida in the Islamic Maghreb, as do most like-minded militants in the region, the official said, speaking on condition of anonymity because he was not authorized to discuss the investigation publicly.

Tunisian Interior Ministry spokesman Tarrouch Khaled confirmed that Harzi was in custody in Tunis. Khaled said "his case is in the hands of justice," but he would not elaborate further.

First human heart cell model of an inherited heart muscle disorder developed

ScienceDaily (Oct. 24, 2012) ? Researchers at the National Heart Centre Singapore (NHCS) have successfully created a human heart cell model of arrhythmogenic right ventricular cardiomyopathy (ARVC), an inherited heart muscle disorder which puts one at high risk of developing life-threatening arrhythmias and sudden cardiac death. The NHCS research team discovered that key characteristics of the disease, such as abnormal "fatty changes" and altered distribution of proteins involved in cell-cell connections (called desmosomal proteins) are reproduced in the heart cells. This novel cellular model for studying the disease could help to improve understanding on how these mutations lead to arrhythmias and clinical manifestations of ARVC.

The study, the first of its kind in the world, was published in the European Heart Journal in July 2012.

The human heart cell model was developed using patient-specific induced pluripotent stem cell (iPSC) technology which converts skin samples from an ARVC patient into heart muscle cells on a petri dish outside the body. This technique is based on the revolutionary iPSC technology of transforming skin cells into stem cells, developed by Professor Shinya Yamanaka, winner of the 2012 Nobel Prize in Physiology/Medicine. The NHCS research team has taken a step further by developing a key clinical application of the iPSC technology by replicating one's own heart cells outside the body for the study of genetic cardiovascular diseases.

Associate Professor Philip Wong, Director, Research and Development Unit, NHCS said, "For the first time, we have created a 'crystal ball' of the disease outside the body, to look into the patient's detailed genetic makeup and its relationship to the manifestation of disease. There would be significant opportunities now to safely study the effects of environmental factors and treatments, including gene and drug therapy, on such diseases as they do not have to be tested on patients in the first instance."

Genetic mutations in ARVC typically affect the function of desmosomes, which are structures that attach heart muscle cells to one another. Desmosomes provide strength to the heart muscle and play a signalling role between neighbouring cells. Without normal desmosomes, the heart muscle cells will detach from one another and die, particularly when the heart muscle is placed under stress (such as during vigorous exercise). The damaged heart muscle is gradually replaced by fat and scar tissue. These changes also disrupt the electrical signals that control the heartbeat, which can lead to dangerous arrhythmia and sudden cardiac death.

ARVC occurs in an estimated 1 in 2,000 to 1 in 5,000 people. The disorder may be under-diagnosed as it can be difficult to detect in people with mild or no symptoms. "Although ARVC is a rare condition, it is more commonly detected in younger individuals, in their 20s and 30s, particularly in males, and is more lethal in this age group," said Dr Reginald Liew, Deputy Director, Research and Development Unit, NHCS and principal investigator of the study. Dr Liew is also an Assistant Professor with the Duke-NUS Graduate Medical School Singapore (Duke-NUS). ARVC may not have any symptoms especially in the early stages. Common symptoms if they do occur include palpitations, light-headedness, and fainting. Those with family history of sudden cardiac death are at higher risk.

The team has also been successful in using the iPSC technology to replicate other inherited heart rhythm diseases such as long QT syndrome (LQTS) and Brugada Syndrome. These diseases are caused by mutations in genes coding for proteins that control the electrical activity of the heart which can lead to ventricular arrhythmias, blackouts and sudden cardiac death.

"Our success in using iPSCs as a platform for the study of genetic cardiovascular diseases was made possible with the clear and cohesive networking between the research scientists, clinicians and bio-medical engineers. The collaborative efforts with other leading research institutions in Singapore, including Duke-NUS, NUS and A*STAR, has allowed NHCS to take a lead globally in this area and enhance Singapore's reputation as a leader in translational cardiovascular research," said Associate Professor Wong. The recognition has been followed with invitations for collaboration from other internationally recognised research centres working in this area.

The 10-member research team comprises six research scientists, two clinician scientists and two staff from the Electron Microscopy Unit, Yong Loo Lin School of Medicine, National University of Singapore. The three-year project which started in 2010, was supported with a research grant from Goh Foundation and administered through Duke-NUS.

"The next stage is for us to use this ARVC model to understand more about the disease and to specifically use such models to risk stratify patients with risk of cardiac arrhythmias. Such models will allow us to measure risk in individuals safely and tailor individual preventive programmes and treatments to patients in a more precise manner, i.e. the practice of 'stratified and personalised' medicine," said Associate Professor Wong.

An ex-convict is in custody after a police officer in Nassau County, N.Y., was killed on the job on Tuesday and a driver was carjacked and shot to death later in the day.

The officer's death came one day after his colleagues attended the funeral of another officer in the police department on?Long Island, reported NBCNewYork.com.

The suspect, Darrel Fuller of Brooklyn, previously served five years in prison for a 2004 attempted murder, according to?The New York Daily News.

Officer?Arthur Lopez, 29, an eight-year police veteran, was killed Tuesday around 11 a.m.?after getting shot in what started out as a traffic stop, reported The New York Daily News. Fuller, 33, was arrested on charges of shooting him as well as another driver, 52-year-old Queens resident Raymond Facey, in a subsequent carjacking, said NBCNewYork.com. Both Officer Lopez and Facey died from their wounds.

"Someone is chasing me," Facey reportedly told his daughter over the phone before Fuller stole his car, reported The Daily News.?

Lopez's death came the day after officers attended the funeral of Nassau County highway patrolman Joseph Olivieri, who was killed last Thursday when he was hit by a car at a crash scene where he had responded.

Lopez, was on patrol Tuesday in Bellerose Terrace, a community at the border of Queens, N.Y., and Nassau County. The officer and his partner spotted a damaged silver Honda that was "running on rims," suspecting it was wanted for leaving the scene of a hit-and-run accident, authorities said, according to NBCNewYork.com.

The officers gave chase, and the car pulled over. There was "a brief exchange of words" between Lopez and the driver before the driver left the vehicle and fired one round into Lopez's chest, said Nassau County Police Department Chief Steven Skrynecki, according to NBCNewYork.com, adding that the officer wasn't wearing a bulletproof vest.

The second officer administered first aid to Lopez, officials said. He was later hospitalized for trauma.

Lopez "lost his life to a cold-blooded killer," Nassau County Executive Edward Mangano said at a news conference.

The shooter got back into his car and drove away before abandoning the vehicle and accosting Facey, the Queens resident, authorities said. He shot the Queens man in the head, dragged him out of his car and left him dead on the roadway as he drove away, they said. That car later was found abandoned in a residential neighborhood in Queens.

The slayings touched off a search for Fuller that included heavily armed NYPD officers going door-to-door in Queens, reported NBCNewYork.com. Authorities also closed the Cross Island Parkway for several hours, causing traffic delays.

Suspect wounded
A 911 caller reported hearing gunfire in Queens amid the massive manhunt for Fuller. Responding officers found him in a vehicle ? not the one carjacked ? with what appeared to be a self-inflicted gunshot wound to the shoulder, said The Daily News.

Fuller was hospitalized in police custody. It was unclear if he had a lawyer.

Fuller pleaded guilty to attempted murder in 2005, reported NBCNewYork.com. He was jailed again in 2010 after violating his parole, but was released in May 2011.

"We're endeavoring to find out what triggered this entire series of events," Skrynecki said while announcing the arrest late Tuesday.

Before the shootings, Fuller left a hospital and had gotten in a car accident, the chief added, but wouldn't elaborate.

A man who witnessed the officer's shooting as he was stopped at a gas station along the highway said it looked like a routine traffic stop.

"The officer's walking up to the car. They just pulled out (a gun) and shot," said Paul Walcott, a music producer from Bayside, N.Y., reported NBCNewYork.com. "He went right down. He got hit point blank. He went straight down."

The car then sped off, heading south on the Cross Island Parkway.

"In broad daylight, this time of day, it was incredible to see something like that," Walcott said.

HELP: Windows Media Center not getting signal

My TV cable comes into my room, goes through a splitter, one goes to my digital converter then to my TV. The other goes to my Windows 7 Desktop.

When I got up Monday evening, Windows Media Center was working fine and it recorded a program that I had set it to record. When I got up Tuesday evening, I discovered it wasn't getting a signal.

I tried switching the outputs on the splitter. TV still got signal, WMC didn't. That rules out a bad splitter. I took the TV cable loose from the back of my Desktop and plugged it into the inputs on both the digital converter and my TV. TV got signal both times. That rules out a bad cable.

I also tried connecting the cable to my Desktop to the output on the digital converter to see what would happen. WMC still did not have a signal. I do not have the means to establish a direct connection to the cable coming into my room to my desktop, but based on the above, I doubt that would work.

I even tried running a system restore to a point before the last known time it was working. Still no luck.

My desktop hasn't moved from the spot it's in since I first turned it on. I plan to disconnect everything, take out outside and blow dust out of it at the end of every month, but I have not had it that long yet.

I tried telling it to scan for more channels twice. Both times got no additional channels found. Then I told it to Set Up TV Signal. Everything went just like it did when I set it up the first time. It successfully downloaded "Latest TV setup data for your region," had no problem examining my TV signal and detected "Digital Cable (ClearQAM) (1 tuner)". I selected configure with these results and clicked next. I selected Maynardville Comcast Maynardville (Digital). Don't know why that came up since I'm much closer to Pigeon Forge and Sevierville, but it worked the first time, so I selected it again. I confirmed my selection. It scanned for TV cannels I receive. It said no channels were detected.

I went throught it again, except instead of configure with these results, I selected "let me configure my TV signal manually." For TV signal type, I selected Cable. Set top box not required by my cable provider, so I selected No. At that point I remembered that the first time, I selected configure with these results, that didn't work and it worked after doing it manually. Moving on, my provider's signal is digital, so like before, I selected Digital Cable (ClearQAM). I selected Maynardville Comcast Maynardville (Digital). It immediately said WMC has finished setting up your Digital Cable (ClearQAM) and asked if I wanted to set up for another TV signal, I selected No. I got The following TV signals will be configured for WMC Digital Cable (ClearQAM) Maynardville Comcast Maynardville (Digital) (1 tuner). Guide download completed immediately and scanning for channels I receive started running.

It said no channels were detected. I restarted my system. I opened WMC and went to the guide. I got an error message saying that all channels had been removed from the guide and to select Edit to add channels to the guide. I also found that all of my scheduled recordings had been removed. I went throught the whole setup thing again. I got no channels were found.

Has anyone had a similar problem? What can I do to diagnose and fix the problem?

>> reporter: good morning. this is such a fascinating story. the whale spent most of his life amusing an amazing marine biologists , and the team has spent decades studying him saying this whale was definitely different than the rest. these sounds may not seem clear to a lot of people, but to some marine biologists what you're hearing is a whale mimicking the human voice . the star of this research project was a beautiful white whale named noisy and he was found in manitoba, canada and later flown to the national marine mammal center for a study. the study team first began to notice human-like sounds in the dolphin and whale enclosure pack in 1984 , sounds they soon figured out had come from the whale .

>> he was the only one who did it to lower the pitch of his voice quite drastically. it was my impression and that of just about everybody that heard it that he was trying to imitate the human voice .

>> reporter: convinced they had no ordinary whale in the study group ridgeway and his group built a special underwater listening platform to capture the sounds and voices coming from the whale .

Brig. Gen. Wissam al-Hassan was killed along with seven others Friday in an attack that many blame on Syria. He was in charge of an investigation over the summer that led to the arrest of a Syrian-allied Lebanese politician over what authorities called a plot by Damascus to provoke chaos with bombings and assassinations.

The probe was an embarrassing blow to Syria, which has long acted with impunity in Lebanon.

Giant posters of al-Hassan were set up around Beirut ahead of Sunday's funeral calling him a "martyr of sovereignty and independence."

With billions of dollars in credit card fraud every year, shoppers may be looking for more secure ways of doing their business. One such way is to use ?temporary? credit card numbers.

They?re sometimes referred to as ?virtual? or ?disposable? numbers. They work in a variety of ways; while not foolproof, they may help consumers to head off abuse or misuse of their credit cards.

Most issuers of major credit cards will let you use a number (not the one on your plastic card) temporarily, often for just a single purchase. After that transaction is complete, the number becomes worthless to a thief or anyone else. The number is truly disposable, so even if a thief gets access to it, the number can?t be used for unauthorized purchases.

You use that number as you would your regular credit card, and your transactions show up on your regular credit card bill. Just as you save receipts from your regular credit card purchases, you should hang onto receipts from any temporary numbers you use.

Every issuer has its own rules for issuing temporary numbers; some tie the service to their online banking and may require you to sign up for it. Others offer a downloadable program that will pop up when you make an online purchase and ask if you want to use a virtual number. Still others require that you log onto their website when you want a virtual number.

What you should not do is click on anything in an unsolicited email offering you a temporary number. That?s likely a phishing scam, designed to harvest your personal information. Since the whole point is to keep that information away from the bad guys, don?t give it away to someone you don?t know. Also, when dealing online, make sure the websites you visit are the ones that truly match your intended destination, not some crook?s computer. Type in the address yourself, or use a bookmark if you?ve made one, rather than clicking on a look-alike link.

You also can request to use a disposable number more than once, and this is where things can get a bit tricky. The practice may give nervous consumers some reassurance, but they can?t defeat all fraud; as long as a disposable number is active, thieves might still make bogus charges with it. Some experts advise using only one-time numbers and that each number applies to a specific merchant.

When sociologist Mike Tomlinson began combing through the health records of people in Northern Ireland, he wasn?t interested in suicide. He was on the hunt for links between poverty and international conflict. But he came across a startling trend. From 1998 to 2008, the rate at which men in their mid-30s to mid-50s were committing suicide rose alarmingly fast, more quickly than the rate for the rest of Northern Ireland?s population.

At first, that spike made no sense. A peace agreement reached in 1998 transformed Northern Ireland into a prosperous and tranquil place. Economic indicators had been surprisingly good. Suicide rates in neighboring countries were all gently falling. Nothing seemed to explain why so many of these men were killing themselves.

But Tomlinson found a hint in the men?s pasts. They had all grown up in the late 1960s and the 1970s, during some of the worst violence Northern Ireland had ever experienced. Called the Troubles, this warlike period brought religious and political fighting that pitted neighbor against neighbor. Children of the Troubles lived with terrorism, house-to-house searches, curfews and bomb explosions. Trauma early in life had rendered men more vulnerable to taking their own lives later, Tomlinson proposed in July in International Sociology.

?If you were younger then, you carry that through,? says Tomlinson, of Queen?s University Belfast. This idea, that something that happened long ago could have such a profound effect today, seemed to resonate with others. When he described his idea to a suicide prevention group in Northern Ireland, ?they just lit on it, and said it speaks so much to what they were seeing.?

Tomlinson does not study the brain, but his work has led him to an idea that?s been under close scrutiny by people who do. Neuroscientists and psychologists now believe that childhood trauma, including violence and neglect, sears itself into the brain in ways that can have devastating effects later.

?It?s a known fact that individuals with early life adversity are at a higher risk of suicide,? says Gustavo Turecki, who directs the McGill Group for Suicide Studies in Montreal. A 2001 study in the Journal of the American Medical Association looking at more than 17,000 Californians found that harmful childhood experiences boost a person?s lifetime risk of attempting suicide by two to five times. Other studies reveal that people who experienced adversity during childhood make up anywhere from 10 to 40 percent of people who later display suicidal behavior.

All the evidence suggests that childhood trauma can lead to suicide. Now, Turecki says, scientists have to figure out why.

In the last few years, they have begun to turn up molecular scars from past abuse. Some researchers have discovered chemical tags that change genes? behavior in ways that may contribute to suicide. And new evidence reveals that childhood trauma may throw off-kilter the hardware responsible for the brain?s response to stress. For a person struggling with suicidal thoughts, dealing with stress appropriately can be a matter of life or death. As more and more signs of these brain changes turn up, the scientific community is struggling to understand how they affect a person?s actions, thoughts and decisions ? behaviors that sway a person?s likelihood of committing suicide.

?This is something we see in the clinic,? Turecki says. ?People exposed to traumatic events seem to have a harder time adapting to life.?

The immediate goal, researchers say, is to better identify people who are likely to act on suicidal thoughts. But in the future, a deeper understanding of the brain scars left by abuse might point to better treatments for someone battling against suicidal behavior, and perhaps even ways to prevent such tragedy.

Something ?truly biological?

Worldwide, about 1 million people take their own lives each year. And the number appears to be growing: In the last 45 years, the suicide rate has jumped by 60 percent.

Every suicide is different. But when scrambling to explain why people would take their own lives, researchers look for similarities, any common strands that might make some sense of the senseless.

Men are more likely to die from suicide than women, though women are more likely to attempt suicide, the stats show. Caucasians are more likely to commit suicide than African Americans. Smokers, substance abusers and people with extremely low cholesterol are all more likely to die by suicide than their counterparts. Genetics undoubtedly plays a role, because suicides are known to run in families. The chemical messenger serotonin, best known for its job in regulating mood, also seems to have a part: People who commit suicide are thought to have dampened serotonin signaling.

But no one knows whether these factors are causes of suicidal behavior or just innocent bystanders that happen to show up commonly in people who commit suicide. That distinction isn?t clear even for two of the strongest suicide predictors.

For one, most people who commit suicide are in the throes of an unmanaged psychiatric illness. And another, many of these people have recently experienced something very stressful, like public humiliation or a family problem, some precipitating factor that seems to send them over the edge. But attributing suicide solely to a stressor, or solely to a mental illness, is an oversimplified view, says psychiatrist J. John Mann of Columbia University and the New York State Psychiatric Institute in New York City. There must be much more to the story.

?What you see ? and this is what the press gets locked into without looking beyond it ? is an immediate precipitant. Job problems, scandal in the government, those kinds of things,? Mann says. ?But in fact, people are dealing with psychological stressors all the time, and compared to that, the number of people who die by suicide is tiny. Why doesn?t that happen all the time if it?s cause and effect??

Something different, above and beyond a mental illness or an intensely stressful situation, is present in the brains of people who commit suicide, Mann contends. ?Something truly biological is happening,? he says.

Evidence now suggests that, in some cases, that something originates in traumatic early life experiences. The thread leading from a rough upbringing to suicide is quite strong.

Altered stress

Rats aren?t known to commit suicide, but a poor upbringing does have profound effects on their brains. Rats neglected by their mothers have brain changes that cause the animals to grow up with abnormal responses to stress, Michael Meaney of McGill University and his team found in a series of experiments in the 1990s. Turecki was intrigued by Meaney?s results. ?We thought the same mechanism might make sense in humans,? Turecki says.

They turned out to be right. Turecki, Meaney and colleagues have now found changes in people similar to those seen in rats. And misbehaving genes in the brain seem to be behind both results.

By amassing a large collection of brain tissues from people who committed suicide, the researchers have been able to study genes, proteins and structures to look for similarities. One particular gene, found to behave abnormally in the neglected rats, caught the researchers? interest. This gene?s activity level is low in people who suffered childhood adversity and later committed suicide, the team found. This wasn?t true for people who had normal childhoods and committed suicide or for people who died in accidents.

The gene in question makes a protein, called the glucocorticoid receptor, that decides when the body?s stress system has produced enough of the stress-signaling hormone cortisol and helps shut that system down. Without enough of this receptor, the body and brain can?t reset after a stressful event.

In people who committed suicide who also suffered childhood abuse, a chemical stop sign was affixed onto the gene in a certain part of the brain, Turecki and his team reported in Nature Neuroscience in 2009. This stop sign, presumably attached during childhood, might stymie current and future production of the glucocorticoid receptor. (The team has no way of knowing the timing for sure because they can study the brain only once a person has died.) Since publishing that finding, the researchers have turned up further evidence: This stop signal, a chemical tag called a methyl group, is also present on several different parts of the gene that encodes the glucocorticoid receptor, Turecki and colleagues reported July 1 in Biological Psychiatry.

And the sensitivity of glucocorticoid receptor production to trauma seems to start early. Karl Radtke of the University of Konstanz in Germany and colleagues found similar methyl tags on the gene in blood samples of children born to women who experienced domestic violence while pregnant. Though the researchers can?t say for sure what is happening in the children?s brains or when the methyl tags first appeared, the results, reported online last year in Translational Psychiatry, do suggest that trauma in the earliest stages of development can have a lasting influence.

Genetic effects of trauma during childhood aren?t restricted to the gluco?corticoid receptor, either. In a study comparing brain tissue of 25 people who died by suicide and experienced childhood trauma with 16 people who died suddenly without abuse, control regions for more than 300 genes had differing methyl tags, Turecki and colleagues reported in the July Archives of General Psychiatry.

It?s far from clear whether these genes have a role in suicide. Without large samples of people who experienced abuse without suicide, it?s impossible to get at any clear links.

But scientists are starting to figure out where to look next for possible clues.

A faulty axis

The glucocorticoid receptor is one small piece of the body?s larger stress system, which may be molded in many ways by abuse in early childhood. Called the HPA axis, this stress-response unit is made up of three pillars: the hypothalamus and pituitary gland in the brain and the adrenal glands on the kidneys. In response to a stressful situation, the HPA axis kicks into gear, churning out stress hormones that help a person get through challenging times. During childhood, this system is shaped by the environment. As children grow up, the HPA axis hardens into its final form. Some researchers, including Turecki, believe that a stress-response system permanently altered by early life trauma could lead to suicide later.

?To some degree, the message that you?re getting is that the environment is hostile,? Turecki says. ?You?re being abused by people you?re supposed to trust, people who develop relationships of trust and attachment. When they?re abusing you, the message you?re getting is the world is one in which you can?t trust anyone. You?re always on alert.?

Many studies, both in animals and in people, have found that adversity early in life programs this stress system for a quick ramp-up, like an engine that roars to life at the lightest tap on the accelerator. What?s more, the engine keeps the RPMs redlined longer than it should.

Exactly how this system is sculpted by adversity in childhood is still unclear, but some animal experiments have yielded good leads. Changes in the structure and function of a brain region called the hippo?campus, which is known to be sensitive to the kind of stress experienced during trauma and is also involved in controlling the stress response, seem to be involved. Levels of molecular players that fluctuate along with stress, such as the protein BDNF and the chemical messenger glutamate, could also be disrupted. And some results hint that early stressors can even staunch the production of new nerve cells, which may somehow preclude a normal stress response later.

Jussi Jokinen of the Karolinska Institute in Stockholm and colleague Peter Nordstr?m have been turning up evidence for HPA axis malfunctioning in people at risk of suicide. Recently, the team has found that a stress response, measured by cortisol in the blood, lingers too long in young adults with mood disorders such as depression who have attempted suicide, compared with others with mood disorders who have not attempted suicide. Another study has found that the cortisol-producing adrenal glands weigh more in people who committed suicide, suggesting that the enlarged organs have adapted to pumping out massive amounts of stress hormones.

From brain to behavior

As researchers move forward, the real challenge will be to identify the complex behaviors that may be the outward sign of suicide-associated brain changes.

Certain personality traits, such as anxiety, aggression, impulsivity and poor decision-making, show up in people exposed to childhood adversity. ?We know there are alterations in executive function in kids who have been neglected,? says child psychiatrist David Brent of the University of Pittsburgh School of Medicine.

And such traits also appear in people who commit or consider committing suicide. Brent and his colleagues have recently found poor decision-making in adolescents who have previously attempted suicide. In a classic lab gambling task in which people picked cards from different decks, these volunteers kept choosing to draw from the losing deck of cards long after other people identified the winning deck. The study, led by Jeff Bridge of the Nationwide Children?s Hospital in Columbus, Ohio, was published in the April Journal of the American Academy of Child and Adolescent Psychiatry.

?What we have are different pieces of the puzzle,? Turecki says. It?s too early to draw clean lines from a hard childhood to a changed brain to behavior and on to suicide. And even if the relationship between early life and suicide were clear, the effect wouldn?t apply to everyone. But the results, though incomplete, are starting to paint a compelling picture that could explain some ? not all ? suicides.

Although there are many loose ends, the work so far suggests that in some cases, early childhood adversity changes the brain in a way that primes it for self-destructive behavior. Bigger studies of more diverse groups of people will help reveal how all these diverse lines of data fit together, says Brent. ?These strands converge,? he says. ?They converge on a person?s ability to balance the will to live and the wish to die.?

Often, when people come into a clinic or emergency room with signs of suicidal behavior, there?s no good way to figure out whether those people are truly a risk to themselves or not. With a deeper understanding of how brain changes early in life can usher in a greater risk of suicide, doctors may be better able to better judge whose life is in danger. Particular behaviors and ways of thinking might be good outward signs of what?s happening inside the brain, giving doctors clues about who needs the most help. ?This is a story that?s just beginning,? Turecki says. ?There?s so much still to learn.??


Marked early? Researchers have identified a number of molecules that may play a role in suicide or related mental problems. Amounts of these molecules in the body can be modified by life experiences and environment, often via chemical tags that affix to DNA and influence how a gene behaves.?

BDNF? The BDNF protein, which helps brain cells grow and survive, is diminished in people who commit suicide. Chemical tags called methyl groups may influence the activity of the gene for BDNF, slowing protein production.

TrkB? Low levels of this protein, which works with BDNF in the brain, have been linked to suicide. A DNA region that controls the gene for TrkB has more methyl tags in the brains of people who commit suicide.?

Polyamines? In people who commit suicide, gene activity controlling levels of polyamines ? small molecules that help regulate cell growth and influence cell-to-cell communication in the brain ? appears to be altered.

5-HTT? This protein slurps up serotonin, which helps nerve cells communicate and has been linked to suicide. A DNA region controlling activity of the gene for 5-HTT had more methyl tags in abused children than in others.
